Our Comments:

We're going to have to assume that by 'prepared' you meant that it was prepared using powdered mustard with other ingredients. Dijon mustard is a prepared mustard. Ballpark mustard is another type of mustard.

We're also going to assume that by 'regular mustard' you meant that the mustard was the yellow kind that typically comes in a bottle.

Having corralled all of our assumptions, we're going to go out on a limb and say, yes, you can substitute prepared for regular. In fact, we personally know two people who won't eat regular mustard.
